1.首要(yao)原则:细(xi)(xi)(xi)胞(bao)不重(zhong)要(yao)情况下(xia)立即(ji)丢弃,培(pei)养(yang)(yang)(yang)箱灭(mie)菌,所(suo)用(yong)(yong)培(pei)养(yang)(yang)(yang)基(ji)也都(dou)(dou)(dou)(dou)要(yao)丢弃,器械(xie)等重(zhong)新灭(mie)菌或(huo)拆(chai)用(yong)(yong)新的(de)。2.细(xi)(xi)(xi)菌污(wu)染(ran)一(yi)般都(dou)(dou)(dou)(dou)救不回来了,发(fa)现(xian)的(de)时(shi)(shi)(shi)候(hou)培(pei)养(yang)(yang)(yang)基(ji)一(yi)般都(dou)(dou)(dou)(dou)很浑浊且细(xi)(xi)(xi)胞(bao)都(dou)(dou)(dou)(dou)死了3.污(wu)染(ran)且细(xi)(xi)(xi)胞(bao)很重(zhong)要(yao)时(shi)(shi)(shi):遇(yu)到念球菌污(wu)染(ran),且细(xi)(xi)(xi)胞(bao)为基(ji)因改造细(xi)(xi)(xi)胞(bao),非常(chang)重(zhong)要(yao)。如231贴壁(bi)乳(ru)腺(xian)细(xi)(xi)(xi)胞(bao),发(fa)现(xian)细(xi)(xi)(xi)胞(bao)周围出现(xian)很小的(de)串珠透亮圆(yuan)点,非常(chang)像念球菌污(wu)染(ran),此(ci)时(shi)(shi)(shi)细(xi)(xi)(xi)胞(bao)状态(tai)尚可,且污(wu)染(ran)少。处理如下(xia):用(yong)(yong)预热或(huo)室(shi)温PBS清(qing)洗(xi)(xi)3次(ci),可适当振(zhen)摇,将(jiang)污(wu)染(ran)冲洗(xi)(xi)下(xia)来。随(sui)后(hou)(hou)(hou)加入10-20%双抗到培(pei)养(yang)(yang)(yang)瓶,置(zhi)于37度培(pei)养(yang)(yang)(yang)箱1h,之(zhi)后(hou)(hou)(hou)再用(yong)(yong)PBS清(qing)洗(xi)(xi)三遍(bian),直至视野下(xia)无可见污(wu)染(ran)。此(ci)时(shi)(shi)(shi)细(xi)(xi)(xi)胞(bao)也被冲下(xia)大部分(fen),因此(ci)此(ci)方法只适用(yong)(yong)在细(xi)(xi)(xi)胞(bao)贴壁(bi)强,状态(tai)好,密度高时(shi)(shi)(shi)使用(yong)(yong)。之(zhi)后(hou)(hou)(hou)每(mei)天再更换培(pei)养(yang)(yang)(yang)基(ji),每(mei)次(ci)用(yong)(yong)PBS冲洗(xi)(xi)2遍(bian)。过几天细(xi)(xi)(xi)胞(bao)状态(tai)尚可时(shi)(shi)(shi),消化离心时(shi)(shi)(shi)用(yong)(yong)500r,3min,去掉(diao)上清(qing),重(zhong)复3次(ci)。这个方法是(shi)根据文(wen)献可利用(yong)(yong)念球菌和细(xi)(xi)(xi)胞(bao)体积重(zhong)量差异(yi)实现(xian)分(fen)离。基(ji)本(ben)上这一(yi)步做完(wan)以后(hou)(hou)(hou),污(wu)染(ran)就基(ji)本(ben)了,接下(xia)来就注意(yi)多(duo)观察,勤换液就行。细(xi)(xi)(xi)胞(bao)增殖与细(xi)(xi)(xi)胞(bao)凋(diao)亡、细(xi)(xi)(xi)胞(bao)周期等是(shi)研究(jiu)(jiu)的(de)重(zhong)要(yao)表型,是(shi)分(fen)子生物学和药理学研究(jiu)(jiu)解决(jue)的(de)问题(ti)之(zhi)一(yi)。广西组(zu)织科研技(ji)术服(fu)务实验室(shi)
